补阳还五汤对急性脑梗塞再灌注大鼠心肌损伤标志物的影响

摘要:目的:探讨补阳还五汤预处理对急性脑梗死再灌注大鼠心肌损伤标志物肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)、肌钙蛋白Ⅰ(cTnⅠ)的影响。方法:将48只SD大鼠随机分为补阳还五汤高剂量组、补阳还五汤低剂量组、模型组、假手术组各12只。模型组及假手术组灌胃无菌水,补阳还五汤高、低剂量组给予补阳还五汤颗粒剂溶液灌胃,给药剂量分别为29.69g/(kg·d)及14.84g/(kg·d),连续14天。采用大脑中动脉线栓法制作急性脑梗死再灌注模型,ELISA法检测血清心肌损伤标志物CK、CKMB、cTnⅠ水平。结果:与假手术组比较,模型组大鼠血清CK、CK-MB含量显著升高,差异均有统计学意义(P<0.01),说明模型成立。与模型组比较,补阳还五汤高、低剂量组大鼠血清CK、CK-MB含量显著降低,差异均有统计学意义(P<0.05,P<0.01)。结论:急性脑梗死再灌注可引起血清心肌酶CK、CK-MB升高,补阳还五汤预处理可降低其CK、CK-MB含量,提示补阳还五汤对急性脑梗死再灌注引起的继发性心肌损伤有保护作用。

Abstract:Abstract:Objective:To investigate the protective effect of Buyang Huanwu tang on myocardial injury markers including creatine kinase(CK),creatine kinase isoenzyme(CK-MB)and troponin I(cTn I)in acute cerebral infarction rats given reperfusion.Methods:Divided 48 SPF SD rats into Buyang Huanwu tang high dosage group,Buyang Huanwu tang low dosage group,the model group,and the sham operation group,12 rats in each.The model group and the sham operation group received aseptic water by gavage,while Buyang Huanwu tang high and low dosage groups were respectively given Buyang Huanwu solution of 29.69 g/(kg·d)and 14.84 g/(kg·d)by gavage,14 days continuously.The method of middle cerebral artery occlusion was used to establish the acute cerebral infarction model of reperfusion,and the enzyme-linked immunosorbent assay(ELISA)was used to detect levels of CK,CK-MB and cTn I.Results:Comparing with those in the sham operation group,levels of CK and CK-MB in the model group were significantly increased,differences being significant(P<0.01),which suggested that the model was valid.Comparing with those in the model group,levels of CK and CK-MB in Buyang Huanwu tang high and low dosage groups were obviously decreased,differences being significant(P<0.05,P<0.01).Conclusion:Reperfusion of acute cerebral infarction can increase the levels of CK and CK-MB,and the pretreatment of Buyang Huanwu tang can reduce the levels of CK and CK-MB,implying that Buyang Huanwu tang has protective effect on secondary myocardial injury caused by reperfusion of acute cerebral infarction.
