    目的:探讨花生红衣通过半胱氨酸蛋白酶(Caspase) 途径对化疗后血小板减少症小鼠的影响。方法:将84 只昆明小鼠随机分为正常组、模型组、白介素组、花生红衣低、中、高剂量组(低、中、高剂量组),每组14 只。除正常组外,其余各组通过腹腔注射环磷酰胺法构建血小板减少症小鼠模型。模型制备成功后,低、中、高剂量组分别按5 mL/kg 的剂量灌胃0.25 g/mL、0.50 g/mL、1.00 g/mL 的花生红衣水提液;正常组、模型组灌服等量蒸馏水;白介素组按0.25 μg/g 的剂量皮下注射重组人白细胞介素-11(rhIL-11);每天1 次,共7 d。给药7 d 后,检测各组小鼠血小板水平及半胱氨酸蛋白酶-3(Caspase-3)、半胱氨酸蛋白酶-9(Caspase-9) 表达情况。结果:与正常组比较,模型组小鼠Caspase-3、Caspase-9 蛋白表达水平升高(P<0.05),血小板水平减少(P<0.01)。与模型组比较,中、高剂量组Caspase-3 蛋白表达降低(P<0.05);中剂量组Caspase-9 蛋白表达降低(P<0.05);低、中、高剂量组与白介素组血小板水平升高(P<0.05)。低、中、高剂量组Caspase-3、Caspase-9 蛋白表达及血小板水平与白介素组比较,差异无统计学意义(P>0.05)。结论:花生红衣可通过下调Caspase-3、Caspase-9 蛋白表达,减少血小板的凋亡,升高血小板水平,治疗化疗后引起的血小板减少症。

    Abstract:

    Abstract: Objective: To investigate the effect of peanut skins on mice with thrombocytopenia after chemotherapy through caspase pathways. Methods:A total of 84 Kunming mice were randomly divided into the normal group,the model group,the interleukin group,the low-dose peanut skin group,the medium-dose peanut skin group,and the high-dose peanut skin group, 14 mice in each group. Except the normal group, the mice models with thrombocytopenia were established by intraperitoneal injection of cyclophosphamide. After the successful preparation of the models,the low-dose, medium-dose and high-dose groups were respectively gavaged with 0.25 g/mL,0.50 g/mL and 1.00 g/mL water extracts of peanut skins at a dose of 5 mL/kg;the normal group and the model group were given equal amount of distilled water;the interleukin group received subcutaneous injection of recombinant human interleukin- 11(RHIL- 11) at a dose of 0.25 μg/g; both groups were treated once a day for seven days. After seven days,the levels of platelet and expression of Caspase- 3 and Caspase-9 were measured in each group. Results:Compared with those in the normal group,the expression levels of Caspase- 3 and Caspase- 9 in the model group were increased(P<0.05), the level of platelet was decreased(P<0.01). Compared with that in the model group, the expression levels of Caspase- 3 in the medium- dose and high- dose groups were decreased(P<0.05),the expression level of Caspase- 9 in the medium- dose was decreased(P<0.05),the levels of platelet in the low-dose,medium-dose and high-dose groups and the interleukin group were increased(P<0.05). There was no significant difference in the comparison of the expression levels of Caspase- 3 and Caspase- 9 and the levels of platelet between the the low- dose,medium- dose and high- dose groups and the interleukin group(P >0.05). Conclusion:Peanut skins can reduce the apoptosis of platelet and increase the platelet level to treat thrombocytopenia due to chemotherapy by down-regulating the expression of Caspase-3 and Caspase-9.
