    目的:通过观察慢性萎缩性胃炎(CAG)大鼠胃黏膜腺上皮主细胞、壁细胞超微结构的变化,探讨健脾清热活血中药胃萎清对CAG大鼠胃黏膜超微结构的影响,初步揭示胃萎清对CAG的作用机制。方法:将CAG大鼠随机分为6组:对照组、模型组、胃萎清高、中、低剂量组及叶酸组。以N-甲基-N'-硝基-N'-亚硝基胍(MNNG)合饥饱失常法建立CAG大鼠模型,观察各组大鼠胃黏膜超微结构的变化。结果:与对照组比较,模型组大鼠胃黏膜主细胞核固缩,胞浆中出现大量空泡样变;壁细胞染色质浓缩,线粒体嵴数量减少。与模型组比较,胃萎清高、中剂量组大鼠胃黏膜主细胞核仁结构尚均一,粗面内质网基本整齐;壁细胞核质比较规则,线粒体嵴增多。胃萎清低剂量组大鼠胃黏膜主细胞细胞核核仁形态较模型组规则;壁细胞细胞核形态改变,线粒体数量减少。叶酸组大鼠胃黏膜主细胞细胞核染色质较正常,胞浆出现空泡样变;壁细胞染色质固缩,线粒体嵴数量较模型组多。结论:胃萎清能在一定程度上改善胃黏膜腺上皮主细胞、壁细胞的超微结构,推测胃萎清能改善CAG患者不适症状的机制之一可能是改善胃黏膜超微结构。

    Abstract:

    Objective:To investigate the effect of Weiweiqing in Chinese medicine for strengthening spleen,clearing heat and activating blood on ultrastructure of gastric mucosa in chronic atrophic gastritis(CAG)rats by observing the ultrastructural changes of gastric mucosal glandular epithelial cells and parietal cells in rats with CAG thus to preliminarily reveal the mechanism of Weiweiqing on CAG.Methods:Divided CAG rats into 6 groups randomly:the controlgroup,the modelgroup,the high-dose Weiweiqing group,the medium-dose Weiweiqing group,the low-dose Weiweiqing group and the folic acid group.Established a CAG ratmodelby app lying N-methyl-N'-nitro-N'-nitrosoguanidine(MNNG)and method of irregular diet.And observe the changes of ultrastructure of gastric mucosa in rats.Results:Compared with the control group,the main cells of the gastric mucosa of rats in the model group showed nuclear pyknosis,a large number of vacuolar degeneration in the cytop lasm,chromatin condensation in parietal cells and a reduced number of mitochondrial cristae.Compared with the modelgroup,the nucleolus structure of gastric mucosalmain cells in the high-dose Weiweiqing group and the medium-dose Weiweiqing group were still the same, and the rough endop lasmic reticulumwere basically neat, and the number of mitochondrial cristae in the parietal cells was increased.Compared with the model group,the pattern of nuclear nucleus of gastric mucosalmain cells in rats in the low-dose Weiweiqing group was more regular.The nucleus morphology of parietal cells changed and the number ofmitochondria decreased.The nuclear chromatin of the gastric mucosalmain cells in the folic acid group was more normal,and there was vacuolar degeneration in the cytop lasmand chromatin condensation in the parietal cells.The number of mitochondrial cristae was larger than that in the model group.Conclusion:Weiweiqing can improve the ultrastructures of gastric mucosal glandular epithelial main cells and parietal cells to a certain extent.It is speculated that one of the mechanisms to improve the ultrastructure of gastric mucosa is that Weiweiqing can improve the discomfort of patients with CAG may be.
