Objective:To discussthe effect and mechanism of aqueousextract of millettia speciosa champ for drug-induced liver fibrosis in zebrafish.Methods:Selected a 3-month-old wild-type zebrafish in AB strain as a research object,and made a zebrafish model of drug-induced liver fibrosis by stimulating with diethylnitrosamine(DEN)for 4 weeks,followed by intervention of high-,medium-and low-concentrations of aqueous extract of millettia speciosa champ.Taken liver tissue for pathological HE staining,sirius red picric acid staining, and Collagen I(Collagen I)immunohistochemical staining, detected expression levelsof α-smooth muscle actin(α-SMA), tumor necrosisfactor- α (TNF- α)and Bcl-2-associated X Protein(Bax)in zebrafish liver homogenate.Results:Compared with the model group,the degree of drug-induced liver fibrosis,pathological injury and collagen fiber deposition in each group received administration of aqueous extract of millettia speciosa champ was all reduced to some extent,and the effect was most significant in the group of high-concentration of millettia speciosa champ(100 mg/L);The expressions of Collagen-1 and apoptosis-related genes α-SMA,TNF-α and Bax in the liver of zebrafish in the group of high-concentration of millettia speciosa champ were significantly decreased,difference being significant(P<0.01).Conclusion:The protective effect of aqueous extract of millettia speciosa champ for drug-induced liver fibrosis is achieved by inhibiting hepatocellular apoptosis and reducing collagen fiber deposition.
