Salidroside has Protective Effect on Injured Nerve in Rats after Cerebral Ischemiareperfusion through Regulating the Activity of JAK2/STAT3 Signaling Pathway

目的:探讨红景天苷能否通过调控Janus蛋白酪氨酸激酶2/信号转导及转录激活蛋白3(Janus kinase2/Signal transducer and activator of transcription3,JAK2/STAT3)信号通路活性对脑缺血再灌注大鼠受损神经元起保护作用。方法:将30只成年健康雄性SD大鼠随机分为假手术组、模型组、红景天苷(Sal)组,每组10只。假手术组大鼠不栓塞中脑动脉,并在相同时间腹腔注射等体积生理盐水,模型组大鼠用Longa线栓法建立大鼠中动脉阻塞(MCAO)模型,并在与Sal组治疗相同时间腹腔注射等体积生理盐水,Sal组大鼠在栓塞2 h后立即腹腔注射50 mg/kg的Sal溶液,连续给药6天,每天1次。记录各组大鼠神经功能损伤评分;TUNEL法检测各组大鼠脑组织内细胞凋亡;Western blot法测定大鼠脑组织内JAK2、p-JAK2、STAT3、p-STAT3的蛋白表达;RT-qPCR技术检测大鼠脑组织内B淋巴细胞瘤-2(B-cell lymphoma-2,Bcl-2)mRNA的表达。结果:与假手术组比较,模型组大鼠神经功能损伤评分升高(P<0.05),脑组织内凋亡神经细胞显著增多(P<0.05),p-JAK2、p-STAT3蛋白表达及Bcl-2 mRNA表达显著增加(P<0.05);与模型组比较,Sal组大鼠神经功能损伤评分下降(P<0.05),脑组织内凋亡神经细胞显著减少(P<0.05),p-JAK2、p-STAT3蛋白表达及Bcl-2 mRNA表达显著降低(P<0.05)。结论:Sal能显著改善脑缺血再灌注大鼠神经功能损伤,降低脑组织内神经细胞凋亡,该作用可能是通过调控JAK2/STAT3信号通路活性实现的。
Abstract:
Objective:To discuss whether salidroside(Sal)can protect the injured neurons in rats after cerebral ischemiareperfusion through regulating the activity of janus kinase 2/signal transducer and activator of transcription 3(JAK2/STAT3)signaling pathway.Methods:A total of 30 healthy adult male SD rats were randomly divided into the sham operation group,the model group and the Sal group,10 rats in each group.In the model group and the Sal group,the rat models with middle cerebral artery occlusion(MCAO)were established by Longa's suture-occluded method.The Sal group was given 50 mg/kg of Sal solution by intraperitoneal injection after occlusion for two hours,and the model group and the sham operation group with no middle cerebral artery occlusion received intraperitoneal injection of saline of the same volume at the same time.All groups were given administration once a day for six days.The score of injury of nerve function in each group was recorded.The apoptosis in brain tissue in each group was detected by TUNEL method;the expression of JAK2,p-JAK2,STAT3 and p-STAT3 protein by Western blot method;and the expression of mRNA of B-cell lymphoma-2(Bcl-2)by RP-qPCR.Results:Compared with those in the sham operation group,the score of injury of nerve function in the model group was increased(P< 0.05),the number of apoptotic neurocytes in the brain tissue was significantly increased(P< 0.05),and the expression of p-JAK2 and p-STAT3 protein and mRNA of Bcl-2 was significantly increased(P<0.05).Compared with those in the model group,the score of injury of nerve function in the Sal group was decreased(P < 0.05),the number of apoptotic neurocytes in the brain tissue was significantly decreased(P< 0.05),and the expression of p-JAK2 and p-STAT3 protein and mRNA of Bcl-2 was significantly decreased(P<0.05).Conclusion:Sal can significantly improve the injury of nerve function in rats after cerebral ischemia-reperfusion and reduce neuronal apoptosis in the brain tissue,whose mechanism may be the regulation of the activity of JAK2/STAT3 signaling pathway.
