制首乌游离蒽醌对高脂血症大鼠血脂及SOD、CTSS mRNA 表达的影响

    目的:探讨制首乌游离蒽醌对高脂血症大鼠血脂水平的影响及其作用机制。方法:将SD大鼠分为对照组、造模组,造模组采用高脂饲料喂饲+脂肪乳剂灌胃的方法复制大鼠高脂血症模型,模型制备成功后,将造模组分为模型组、辛伐他汀组、制首乌游离蒽醌低、中、高剂量组(低、中、高剂量组)。各给药组给予相应的药物,模型组和对照组给予生理盐水。干预5周后,股动脉采血检测大鼠血清甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)含量,采用逆转录-聚合酶链式反应(RT-PCR) 及琼脂糖凝胶电泳检测大鼠肝脏组织蛋白酶S(CTSS) mRNA、超氧化物歧化酶-1(SOD1)、超氧化物歧化酶-2(SOD2) mRNA的表达。结果:与对照组比较,模型组TG、TC、LDL-C含量显著升高(P<0.01);与模型组比较,辛伐他汀组及低、中、高剂量组TG、LDL-C含量显著降低(P<0.05),高剂量组TC含量显著降低(P<0.05);与辛伐他汀组比较,高剂量组TC 含量降低(P<0.05),LDL-C 含量升高(P<0.05)。RT-PCR 结果显示,与对照组比较,模型组CTSS mRNA 相对表达量增加,但差异无统计学意义,SOD1、SOD2 mRNA 相对表达量明显降低(P<0.05);与模型组比较,辛伐他汀组及低、中、高剂量组SOD1 mRNA 相对表达量增加(P<0.05),辛伐他汀组及低剂量组SOD2 mRNA 相对表达量增加(P<0.05);与辛伐他汀组比较,高剂量组SOD2 mRNA表达较低(P<0.05)。结论:制首乌游离蒽醌能下调高脂血症大鼠血脂水平。其作用机制可能与上调SOD1、SOD2 mRNA的相对表达有关,而低剂量组上调SOD2 mRNA的相对表达最显著。

    Abstract:

    Abstract: Objective: To discuss the effect of free anthraquinone in Radix Polygoni Multiflori Praeparata cum Succo Glycines Sotae on levels of blood lipid in rats with hyperlipidemia and its mechanism. Methods:SD rats were divided into the control group and the modeling group. In the modeling group,the rat models with hyperlipidemia were made by high fat diet and fat emulsion gavage. After successful establishment, the modeling group was divided into the model group, the simvastatin group and the free anthraquinone in Radix Polygoni Multiflori Praeparata cum Succo Glycines Sotae group of lowdose, medium-dose and high-dose(the low-dose,medium-dose and high-dose groups). Each administration group was given corresponding medicines, and the model group and the control group were given saline. After intervention for five weeks,the content of triglyceride(TG),total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C) and low-density lipoprotein cholesterol(LDL- C) in serum in rats was detected by blood sampling from femoral artery,and the expression of cathepsin S(CTSS), superoxide dismutase- 1(SOD1) and superoxide dismutase- 2(SOD2) mRNA in the liver in rats was measured by reverse transcription polymerase chain reaction(RT-PCR) and agarose gel electrophoresis. Results:Compared with that in the control group,the content of TG,TC and LDL- C in the model group was significantly increased(P<0.01). Compared with that in the model group,the content of TG and LDL-C in the simvastatin group and the low-dose,mediumdose and high- dose groups was significantly decreased(P<0.05), and the content of TC in the high- dose group was significantly decreased(P<0.05). Compared with that in the simvastatin group,the content of TC in the high-dose group was decreased(P<0.05), the content of LDL- C was increased. The result of RT- PCR showed that compared with that in the control group,the relative expression of CTSS mRNA in the model group was increased,but there being no significance in the difference,and the relative expression of SOD1 and SOD2 mRNA was significantly decreased(P<0.05);compared with that in the model group,the relative expression of SOD1 mRNA in the simvastatin group and the low-dose,medium-dose and high-dose groups was increased(P<0.05),and the relative expression of SOD2 mRNA in the simvastatin group and the low-dose group was increased(P<0.05);compared with that in the simvastatin group,the expression of SOD2 mRNA in the high- dose group was lower(P<0.05). Conclusion: Free anthraquinone in Radix Polygoni Multiflori Praeparata cum Succo Glycines Sotae can down-regulate the levels of blood lipid in rats with hyperlipidemia. Its mechanism may be related to the up-regulation of the relative expression of SOD1 and SOD2 mRNA,and the up-regulation of the expression of SOD2 mRNA is the most significant in the low-dose group.
