    目的:探讨逍遥散对慢性应激大鼠海马组织双特异性磷酸酶 14 (DUSP14) mRNA 表达及对 p38 丝裂原活化蛋白激酶(p38 MAPK)、c-Jun 氨基末端蛋白激酶(JNK)、细胞外信号调节蛋白激酶(ERK)、细胞外信号调节蛋白激酶 5(ERK5)等四类丝裂原活化蛋白激酶 (MAPKs) 信号通路活化状态的影响。方法:将 80 只大鼠分为空白组、模型组、氟西汀组及逍遥散组。空白组大鼠常规饲养,每天灌胃 2 mL 生理盐水;模型组大鼠每天灌胃 2 mL 生理盐水后进行慢性应激刺激;氟西汀组和逍遥散组分别灌胃氟西汀和逍遥散后进行慢性应激刺激;剂量均为 10 mL/(kg · d)。检测各组大鼠海马组织 DUSP14 mRNA 表达及p38、磷酸化 p38 (p-p38)、JNK、磷酸化 JNK (p-JNK)、ERK、磷酸化 ERK (p-ERK)、ERK5、磷酸化 ERK5 (p-ERK5) 的相对含量。结果:与空白组比较,模型组 DUSP14 mRNA 表达显著上调 (P<0.01),p-p38/p38、p-JNK/JNK、p-ERK5/ERK5 均显著升高 (P<0.01),p-ERK/ERK 显著降低 (P<0.01)。与模型组比较,逍遥散组和氟西汀组 DUSP14 mRNA 表达均下调 (P< 0.01);逍遥散组 p-p38/p38、p-JNK/JNK 显著降低(P<0.01),p-ERK/ERK 显著升高(P<0.01);氟西汀组 p-p38/p38、p-JNK/JNK、p-ERK5/ERK5 均显著降低 (P<0.01),p-ERK/ERK 显著升高 (P<0.01)。与氟西汀组比较,逍遥散组 p-JNK/JNK、p-ERK5/ERK5 均较高(P<0.05,P<0.01)。结论:逍遥散和氟西汀对慢性应激引起的 DUSP14 mRNA 表达变化及 MAPK 各信号通路活性变化的调节作用趋势基本一致,对 p38 MAPK 信号通路的调节作用相当,但在 ERK5 MAPK 通路中逍遥散未表现出活性调节作用,在 ERK、JNK MAPK 通路活化状态调节过程中氟西汀更显优势。

    Abstract:

    Abstract: Objective: To study the effect of Xiaoyao powder on dual- specificity phosphatase 14(DUSP14) mRNA expressions and four types of mitogen-activated protein kinases(MAPKs) signaling pathways in the activated state,namely p38 mitogen- activated protein kinase(p38 MAPK), c- Jun N terminal protein kinase(JNK), extracellular signal- regulated kinase(ERK) and extracellular signal-regulated kinase 5(ERK5),in hippocampus of chronic stress rats. Methods:A total of 80 rats were divided into the blank group,the model group,the fluoxetine group and the Xiaoyao powder group. The blank group was routinely fed,and given 2 mL of normal saline by gavage every day;the model group was treated with chronic stress stimulation after being given 2 mL of normal saline by gavage every day;the fluoxetine group and the Xiaoyao powder group were treated with chronic stress stimulation after being given 10 mL/(kg · d) of fluoxetine and Xiaoyao powder by gavage respectively. The expressions of DUSP14 mRNA and the relative contents of p38,phosphorylated p38(p-p38),JNK, phosphorylated JNK(p-JNK),ERK,phosphorylated ERK(p-ERK),ERK5 and phosphorylated ERK5(p-ERK5) in hippocampus of rats in each group were detected. Results:Compared with those in the blank group,DUSP14 mRNA expression in the model group was significantly up-regulated(P<0.01),p-p38/p38,p-JNK/JNK,p-ERK5/ERK5 were significantly increased (P<0.01),and p-ERK/ERK was significantly decreased(P<0.01). Compared with those in the model group,DUSP14 mRNA expressions in the Xiaoyao powder group and the fluoxetine group were down- regulated(P<0.01);p-p38/p38 and p-JNK/ JNK in the Xiaoyao powder group were significantly decreased(P<0.01), and p- ERK/ERK was significantly increased(P< 0.01); p-p38/p38 and p-JNK/JNK and p-ERK5 /ERK5 in the fluoxetine group were significantly decreased(P<0.01), and p-ERK/ERK was significantly increased(P<0.01). Compared with those in the fluoxetine group,p- JNK/JNK and p- ERK5/ ERK5 in the Xiaoyao powder group were higher(P<0.05,P<0.01). Conclusion:Xiaoyao powder and fluoxetine have similar regulatory trend on the changes in DUSP14 mRNA expressions and activities of each MAPK signaling pathway caused by chronic stress, and similar regulatory effect on p38 MAPK signaling pathway, but Xiaoyao powder shows no activity regulation in ERK5 MAPK pathway,and fluoxetine shows more advantages in the regulating process of ERK and JNK MAPK pathways in the activated state.
