    摘要:

    目的:探讨苗药复方金荞麦汤治疗慢性非萎缩性胃炎(CNG) 的药效学作用。方法:将60 只大鼠随机分为正常组、模型组、阳性药组及复方金荞麦汤低、中、高剂量组(以下简称JQMⅠ组、JQMⅡ组、JQMⅢ组)。除正常组外,其余各组采用脱氧胆酸钠、酒精、饥饱失常复合方法制备大鼠CNG 模型。JQMⅠ组、JQMⅡ组、JQMⅢ组分别按6.332、12.664、25.328 g/kg(生药) 的剂量灌胃复方金荞麦汤;阳性药组按5.33 g/kg 的剂量灌胃三九胃泰;模型组及正常组大鼠灌胃等体积蒸馏水。观察各 组大鼠胃液、胃黏膜组织超氧化物歧化酶(SOD)、丙二醛(MDA)、血清胃泌素(GAS)、胃蛋白酶活性(PP) 及病理组织学。结果:与正常组比较,模型组大鼠胃液量、游离酸、总酸度、SOD 活性均降低(P<0.05);pH 值、GAS、PP、MDA 含量及胃体部、胃窦部、胃角部炎症积分、病变积分均增高(P<0.05)。与模型组比较,阳性药组大鼠游离酸、总酸度、SOD 活性均升高(P<0.05);胃液量、pH 值、GAS、PP、MDA 含量及胃体部、胃角部炎症积分、病变积分均降低(P<0.05);JQMⅠ、Ⅱ、Ⅲ组大鼠pH 值、GAS、PP 及胃体部、胃角部炎症积分、病变积分均降低(P<0.05);JQMⅠ组、JQMⅢ组SOD 活性升高(P<0.05),MDA 含量降低(P<0.05);JQMⅡ组大鼠胃液量、总酸度升高(P<0.05),MDA 含量降低(P<0.05);JQMⅢ组大鼠胃液量、游离酸、总酸度均升高(P<0.05),胃体部、胃窦部、胃角部炎症积分均降低(P<0.05)。与阳性药组比较,JQMⅠ、Ⅱ、Ⅲ组大鼠胃液量升高(P<0.05);JQMⅠ组大鼠总酸度、GAS、PP 均降低(P<0.05);JQMⅠ组、JQMⅢ组MDA 含量降低(P<0.05);JQMⅡ组SOD 活性降低(P<0.05);JQMⅢ组胃体部、胃窦部、胃角部炎症积分均降低(P<0.05)。与JQMⅠ组比较,JQMⅡ、Ⅲ组大鼠胃液量、GAS、PP 均升高(P<0.05);JQMⅡ组SOD 活性降低(P<0.05)。与JQMⅡ组比较,JQMⅢ组大鼠总酸度、GAS、PP、SOD 活性均升高(P<0.05),MDA 含量及胃体部、胃窦部、胃角部炎症积分降低(P<0.05)。结论:复方金荞麦汤能明显改善复合因素所致的大鼠CNG 胃黏膜损伤,其药效结果支持该方的临床治疗。

    Abstract:

    Abstract:Objective:To discuss the pharmacodynamic effect of Miao nationality's medicine compound Jinqiaomai tang on chronic non-atrophic gastritis(CNG). Methods:A total of 60 rats were randomly divided into the normal group,the model group,the positive medicine group,and the compound Jinqiaomai tang groups of low-dose,medium-dose and high-dose (Group JQMⅠ,Group JQMⅡ,and Group JQM Ⅲ for short). Except for the normal group,in the other groups,CNG rat models were established by sodium deoxycholate,alcohol,and the method of abnormal starvation and overfeeding. Group JQMⅠ,Group JQMⅡ,and Group JQM Ⅲ were respectively given gastric perfusion of compound Jinqiaomai tang at the doses of 6.332,12.664,and 25.328 g/kg(crude medicine). The positive medicine group was given Wei-Tai 999 at the dose of 5.33 g/kg by gavage;the model group and the normal group were given gastric perfusion of distilled water of the same volume. The gastric juice,superoxide dismutase(SOD) and malondialdehyde(MDA) in gastric mucosa tissues,serum gastrin (GAS),the activity of pepsin(PP),and histopathology were observed in each group. Results:Compared with those in the normal group, the volume of gastric juice, free acid, total acidity, and the activity of SOD in the model group were decreased(P<0.05); pH value, the content of GAS, PP and MDA, as well as the scores of inflammation and lesions in gastric body,gastric antrum,and gastric angulus in the model group were increased(P<0.05). Compared with those in the model group,the free acid,total acidity,and the activity of SOD in the positive medicine group were increased(P<0.05); the volume of gastric juice,pH value,the content of GAS,PP and MDA,as well as the scores of inflammation and lesions in gastric body, and gastric angulus in the positive medicine group were decreased(P<0.05). Compared with those in the model group,pH value,the content of GAS and PP,as well as the scores of inflammation and lesions in gastric body,and gastric angulus in Group JQMⅠ,Group JQMⅡ,and Group JQM Ⅲ were decreased(P<0.05);the activity of SOD in Group JQM Ⅰ, and Group JQM Ⅲ was increased(P<0.05), and the volume of MDA was decreased(P<0.05); the volume of gastric juice,and total acidity in Group JQM Ⅱ were increased(P<0.05),and the content of MDA was decreased(P<0.05); the volume of gastric juice, free acid, and total acidity in Group JQM Ⅲ were increased(P<0.05), and the scores of inflammation in gastric body, gastric antrum, and gastric angulus were decreased(P<0.05). Compared with those in the positive medicine group,the volume of gastric juice in Group JQMⅠ,Group JQMⅡ,and Group JQM Ⅲ was increased(P< 0.05);the total acidity,GAS,and PP in Group JQMⅠwere decreased(P<0.05);the MDA contents in Group JQMⅠ,and Group JQM Ⅲ were decreased(P<0.05); the SOD activity in Group JQM Ⅱ was decreased(P<0.05); the scores of inflammation in gastric body,gastric antrum,and gastric angulus in Group JQM Ⅲ were decreased(P<0.05). Compared with those in Group JQMⅠ,the volume of gastric juice,GAS,and PP in Group JQMⅡand Group JQM Ⅲ were increased(P< 0.05);the SOD activity in Group JQMⅡwas decreased(P<0.05). Compared with those in Group JQMⅡ,the total acidity, GAS,PP,and SOD activity in Group JQM Ⅲ were increased(P<0.05);the MDA content and the scores of inflammation in gastric body, gastric antrum, and gastric angulus in Group JQM Ⅲ were decreased(P<0.05). Conclusion: Compound Jinqiaomai tang can effectively improve gastric mucosal lesion induced by compound factors in CNG rats,whose application in clinical treatment is supported by its pharmacodynamic result.
