Abstract:Objective:To discuss the effect of breviscapine on the heart function,oxidative stress,and inflammatory response of rats with acute myocardial infarction(AMI). Methods:A total of 120 SD rats were randomly divided into the sham operation group,the infarction group,the verapamil group,and the verapamil groups of low- dose,medium- dose and high- dose(low- dose, medium- dose and high- dose groups for short), 20 cases in each group. Except for the sham operation group,in the other groups,AMI rat models were established. The verapamil group and low-dose,medium-dose and high- dose groups were given the intraperitoneal injection of the corresponding medicines in terms of corresponding dosage. The sham operation group and the infarction group were given normal saline of the same volume. All groups were treated for four weeks. Left ventricular internal diameter at end- diastole(LVIDd),left ventricular internal diameter at endsystole(LVIDs), fraction shortening(FS), ejection fraction(EF), and stroke volume(SV) of rats were detected by ultrasonic imaging system. The histopathological examination of the heart was conducted by HE staining method. The level of serum myocardial enzyme was measured. The cardiac index(the ratio of the heart weight to body mass) and the percentage of infarction volume were calculated. The activity of antioxidant enzyme and the content of malondialdehyde(MDA) in myocardial tissue were detected. The content of inflammatory cytokines including tumor necrosis factor-α(TNF-α),intercellular adhesion molecule-1(ICAM-1),interleukin-1β(IL-1β),and interleukin-6(IL-6) in myocardial tissue were measured. Results:When compared with those in the sham operation group,in the infarction group,there was disorderly arrangement and rupture of myocardial fiber,decrease of cell number,partial necrosis,and inflammatory cell infiltration in myocardial tissue;LVIDd, LVIDs,the cardiac index,the percentage of infarction volume,the contents of MDA,TNF-α,ICAM-1,IL-1β,and IL-6 in myocardial tissue,as well as the levels of aspartate aminotransferase(AST),creatine phosphokinase(CPK),and lactate dehydrogenase(LDH) in serum were increased(P<0.05, P<0.01); FS, EF, SV, as well as the activity of superoxide dismutase(SOD) and catalase(CAT) in myocardial tissue were decreased(P<0.01). When compared with those in the infarction group,in the low-dose group,the level of LDH in serum and the content of MDA in myocardial tissue were decreased(P< 0.05);in the medium-dose group,high-dose group,and the verapamil group,LVIDs,the cardiac index,the percentage of infarction volume,the levels of AST and LDH in serum,as well as the contents of MDA and ICAM-1 in myocardial tissue were decreased(P<0.05,P<0.01),FS,EF,and SV were increased(P<0.05,P<0.01),and lesion of myocardial tissue was improved with different degrees. When compared with those in the infarction group,in the medium- dose and high dose groups,the level of CPK in serum and the contents of TNF-α,IL-1β,and IL-6 in myocardial tissue were decreased (P<0.05,P<0.01),and the activity of SOD and CAT in myocardial tissue was increased(P<0.05,P<0.01);in the high dose group and the verapamil group,LVIDd was decreased(P<0.05). When compared with those in the verapamil group,in the low-dose group,LVIDd,LVIDs,and the percentage of infarction volume were increased(P<0.05,P<0.01),and FS, EF, and SV were decreased(P<0.01); in the medium- dose group, LVIDd was increased(P<0.01); in the high- dose group,the level of LDH in serum and the content of MDA in myocardial tissue were decreased(P<0.05,P<0.01),and the activity of SOD was increased(P<0.05); in the medium-dose and high-dose groups, the contents of TNF- α,IL-1β, and IL-6 in myocardial tissue were decreased(P<0.05,P<0.01). Conclusion:Breviscapine has significant improving effect on the heart function of rats after AMI,and significant inhibitory effect on oxidative stress injury and inflammatory response after AMI.
