    目的:探讨三七含药血清对L929 成纤维细胞增殖、迁移及血管内皮生长因子(VEGF)、转化生长因子-β(TGF-β)、Ⅰ型胶原蛋白(Collagen Ⅰ) 表达的影响。方法:将28 只SPF 级SD 大鼠随机分为对照组及三七低、中、高剂量组,制备血清。体外培养小鼠成纤维细胞系L929,对照组给予生理盐水血清,低、中、高剂量组给予相应的三七含药血清干预。CCK-8 法检测成纤维细胞增殖,划痕实验检测成纤维细胞迁移,RT-qPCR 及Western Blot 分别检测VEGF、TGF-β、CollagenⅠ的mRNA 及蛋白表达。结果:与对照组比较,低剂量组细胞干预24 h 后OD 值及干预48 h 后迁移愈合百分比升高(P<0.05),VEGF、TGF-β蛋白及mRNA 表达增加(P<0.05);中剂量组细胞干预24 h 后OD 值及干预48 h 后迁移愈合百分比提高(P<0.05),VEGF、TGF-β、CollagenⅠ蛋白及mRNA 表达增加(P<0.05);高剂量组细胞干预24 h 后OD 值及干预48 h 后迁移愈合百分比降低(P<0.05),VEGF、TGF-β 蛋白及mRNA 表达下降(P<0.05)。与低剂量组比较,中剂量组细胞干预24 h 后OD 值及干预48 h后迁移愈合百分比升高(P<0.05),VEGF、TGF-β、CollagenⅠ蛋白及mRNA 表达增加(P<0.05);高剂量组细胞干预24 h 后OD 值及干预48 h 后迁移愈合百分比降低(P<0.05),VEGF、TGF-β 蛋白及mRNA 表达下降(P<0.05)。与中剂量组比较,高剂量组细胞干预24 h 后OD 值及干预48 h 后迁移愈合百分比降低(P<0.05),VEGF、TGF-β、CollagenⅠ蛋白及mRNA 表达下降(P<0.05)。结论:中剂量三七含药血清可促进成纤维细胞增殖和迁移,并上调VEGF、TGF-β、Collagen Ⅰ的表达,且疗效优于低剂量,而高剂量则抑制成纤维细胞增殖、迁移及VEGF、TGF-β 的表达,不利于创伤愈合。

    Abstract:

    Abstract:Objective:To discuss the effect of medicated serum with Radix et rhizoma notoginseng on proliferation and migration of fibroblast L929 as well as expression of vascular endothelial growth factor(VEGF),transforming growth factor-β (TGF-β),and typeⅠcollagen(CollagenⅠ). Methods:A total of 28 cases of SD rats of SPF level were randomly divided into the control group and the Radix et rhizoma notoginseng groups of low,medium and high dose and the serum was prepared. Fibroblast line L929 of mice was cultured in vitro;the control group was given serum with normal saline,and the groups of low,medium and high dose were given corresponding intervention of medicated serum with Radix et rhizoma notoginseng. The CCK- 8 method was applied to detecting proliferation of fibroblast;scratch assay was applied to detecting migration of fibroblast;RT-qPCR and Western Blot was applied to detecting the expression of mRNA and protein of VEGF,TGF-β and Collagen Ⅰrespectivehy. Results: Compared with those in the control group, in the low- dose group, the OD value 24 h after intervention and migration healing percentage 48 h after intervention were increased(P<0.05), and the expression of mRNA and protein of VEGF and TGF- β was increased(P<0.05); in the medium- dose group, the OD value 24 h after intervention and migration healing percentage 48 h after intervention were increased(P<0.05),and the expression of mRNA and protein of VEGF,TGF- β,and Collagen Ⅰwas increased(P<0.05);in the high- dose group,the OD value 24 h after intervention and migration healing percentage 48 h after intervention were decreased(P<0.05),and the expression of mRNA and protein of VEGF and TGF-β was decreased(P<0.05). Compared with those in the low-dose group,in the medium-dose group, the OD value 24 h after intervention and migration healing percentage 48 h after intervention were increased(P< 0.05),and the expression of mRNA and protein of VEGF,TGF-β,and CollagenⅠwas increased(P<0.05);in the high-dose group, the OD value 24 h after intervention and migration healing percentage 48 h after intervention were decreased(P< 0.05),and the expression of mRNA and protein of VEGF and TGF- β was decreased(P<0.05). Compared with those in the medium-dose group,in the high-dose group,the OD value 24 h after intervention and migration healing percentage 48 h after intervention were decreased(P<0.05),and the expression of mRNA and protein of VEGF,TGF-β,and CollagenⅠwas decreased(P < 0.05). Conclusion : Medium-dose medicated serum with Radix et rhizoma notoginseng can promote proliferation and migration of fibroblast,and up-regulate the expression of VEGF,TGF-β,and CollagenⅠ,whose curative effect is better than that of low- dose therapy. High- dose medicated serum with Radix et rhizoma notoginseng inhibits proliferation and migration of fibroblast as well as the expression of VEGF and TGF- β,which is not conducive to healing of wound.
