Abstract:Objective:To discuss the effect of acupuncture and moxibustion at Zusanli point(ST 36) on phospholipase C-γ1 (PLC-γ1) and gastric mucosal cells activity in rats with gastric ulcer. Methods:A total of 30 rats were divided into the normal group,the gastric ulcer group and the intervention group,10 cases in each group. Except for those in the normal group,rat models with gastric ulcer were established in the other two groups,and the intervention group was treated with acupuncture at Zusanli point and so on. The pathology of gastric tissues in rats in all groups were detected by hematoxylin-eosin staining (HE),the inflammatory factors in orbital venous blood of rats by enzyme-linked immunosorbent assay,the apoptosis ratio in gastric mucosa of rats by TUNEL Assay,and the PLCγ-1 expression in gastric tissues of rats by immunohistochemistry. Results:In the normal group,there was no necrosis and inflammatory infiltration in gastric tissues of rats. In the gastric ulcer group, the gastric tissue arrangement was disordered, the ulcer area was locally dilated, and there was hyperplasia of connective tissue and vascular structural disorder. Compared with those in the gastric ulcer group, the inflammation and necrotic parts of the ulcer in the intervention group were relieved,and the ulcer was covered by granulation. Compared with those in the normal group,the levels of interleukin-8(IL-8),tumor necrosis factor-α(TNF-α) in serum and the apoptosis rate of gastric mucosal cells in the gastric ulcer group were increased(P<0.01,P<0.05),and the expressions of PLCγ-1 protein and mRNA in gastric tissues were decreased(P<0.05). Compared with those in the gastric ulcer group,the levels of IL-8 and TNF-α in serum and the apoptosis rate of gastric mucosal cells in the intervention group were decreased(P<0.01, P<0.05), and the expressions of PLCγ- 1 protein and mRNA in gastric tissues were increased(P<0.05). Conclusion: Acupuncture and moxibustion at Zusanli point(ST 36) can reduce the apoptosis of gastric mucosal cells and the levels of inflammatory factors,which may be related to the activation of PLCγ-1 activity.
