Abstract: Objective: To explore the protective effect of Compound Ganduqing on acute liver injury induced by carbon tetrachloride(CCl4) in mice. Methods:A total of 60 Kunming mice were randomly divided into the normal group,the model group,the positive group(biphenyl diester dripping pills) and the Compound Ganduqing groups of high-dose,medium-dose and low- dose. Except for the normal group, the mice models with liver injury in other groups were established by intraperitoneal injection of 0.4% CCl4 olive oil solution. Compound Ganduqing was administered intragastrically at different stages,and then the blood was extracted by the method of eyeball extirpation in the experiment to further obtain serum. The activity of alanine aminotransferase(ALT) and aspartate aminotransferase(AST) in serum was measured,and the content of Fas and interleukin-6(IL-6) mRNA in liver tissue was determined. The pathological changes of liver tissue were observed by microscope. Results:Compared with the normal group,the levels of ALT and AST in serum were up-regulated in the model group(P<0.01),which proved that the model replication was completed. Compared with model group,the activity of ALT and AST in serum in the biphenyl diadipose group and the Compound Ganduqing groups of each dose was decreased(P< 0.01). There was significant difference in the comparison of the expression of Fas and IL-6 mRNA in the liver tissue between the normal group and the model group(P<0.01). Compared with the model group,the expression of Fas and IL-6 mRNA in the liver tissue was decreased in the biphenyl diadipose group and the Compound Ganduqing groups of each dose(P<0.01) (P<0.05). Pathological observation also showed that Compound Ganduqing could significantly improve the liver injury. Conclusion: Compound Ganduqing has significant protective effect on acute liver injury induced by CCl4 in mice, and the specific mechanism of action may be related to the reduction of the expression of related inflammatory factors.
