    摘要:

    目的:观察扶正消瘤颗粒对乳腺癌SKBR-3 细胞HER-2 mRNA 及蛋白表达的影响。方法:将30 只SD 大鼠随机分成扶正消瘤颗粒L 组、M 组、H 组,3 组大鼠分别予1 倍、2 倍、3 倍量的扶正消瘤颗粒灌胃3 d 后,制备扶正消瘤颗粒的大鼠含药血清。体外培养HER-2 阳性的人乳腺癌SKBR-3 细胞, 对数生长期细胞随机取5 组, 分别是空白对照组、赫赛汀(Herceptin) 组及扶正消瘤颗粒的大鼠含药血清低、中、高剂量组(L 组、M 组、H 组),进行相应干预。采用实时PCR 检测HER-2 mRNA 表达,Western Blot 检测HER-2 蛋白表达。结果:扶正消瘤颗粒3 个剂量组及Herceptin 组的HER-2 mRNA 含量与空白对照组比较,在24 h、48 h、72 h 3 个时间点均有下降(P<0.01),提示3 种剂量的大鼠含药血清和Herceptin 均能抑制SKBR-3 细胞的HER-2 mRNA 表达。L 组24 h 和48 h 的HER-2 mRNA 含量及M 组48 h 的HER-2 mRNA 含量与Herceptin 组比较,差异均无统计学意义(P>0.05);L 组72 h,M 组24 h 及72 h,H 组24 h、48 h、72 h 的HER-2 mRNA 表达量与Herceptin 组比较,差异均有统计学意义(P<0.01,P<0.05)。扶正消瘤颗粒3 个剂量组及Herceptin 组24 h 的HER-2 mRNA 含量均最低,48 h 的HER-2 mRNA 含量较24 h 升高,72 h 的mRNA 含量又降低。与空白对照组进行比较,Herceptin 组和L 组的SKBR-3 细胞HER-2 蛋白量均下降,差异均有统计学意义(P<0.01);与Herceptin 组比较,M 组及H 组的HER-2 蛋白量较高,差异均有统计学意义(P<0.01)。L 组的HER-2 蛋白量与Herceptin 组比较,差异无统计学意义(P>0.05)。结论:扶正消瘤颗粒的大鼠含药血清能明显抑制乳腺癌SKBR-3 细胞HER-2 mRNA 表达和HER-2 蛋白表达,其抑制作用较Herceptin 要差一些,扶正消瘤颗粒低剂量的含药血清效果更好。

    Abstract:

    Abstract:Objectives:To observe the effect of Fuzheng Xiaoliu granules on human epidermal growth factor receptor 2 (HER- 2) mRNA and protein expressions in SKBR- 3 cells of breast cancer. Methods: A total of 30 SD rats were randomly divided into the Fuzheng Xiaoliu granules low- dose group(Group L), median- dose group(Group M) and high- dose group (Group H). In the three groups,rats with medicated serum containing Fuzheng Xiaoliu granules were prepared by Fuzheng Xiaoliu granules by gavage at the doses of one, two and three times respectively. HER- 2- positive human breast cancer SKBR-3 cells were cultured in vitro. Rats with cells in the logarithmic growth phase were randomly divided into five groups, with the blank control group, the Herceptin group and the three groups of rats with medicated serum containing Fuzheng Xiaoliu granules at three different doses,and were given corresponding intervention. The HER- 2 mRNA expressions were detected by real-time polymerase chain reaction(PCR),and the HER-2 protein expressions by Western Blot. Results:The HER-2 mRNA contents in the three groups of Fuzheng Xiaoliu granules and the Herceptin group were decreased at 24 h, 48 h and 72 h when compared with those in the blank control group(P<0.01),suggesting that rats with medicated serum at three doses and Herceptin could inhibit the HER- 2 mRNA expressions in SKBR- 3 cells. There was no significant difference being found in the comparison of HER-2 mRNA contents at 24 h and 48 h between Group L and the Herceptin group,and of HER- 2 mRNA contents at 48 h between Group M and the Herceptin group(P>0.05). The comparisons between HER- 2 mRNA expressions in Group L at 72 h, Group M at 24 h and 72 h, Group H at 24 h, 48 h and 72 h and those in the Herceptin group showed significant differences(P<0.01,P<0.05). The HER-2 mRNA contents at 24h in the three groups of Fuzheng Xiaoliu granules at three doses and the Herceptin group were the lowest,the HER-2 mRNA contents at 48 h were increased when compared with those at 24 h,and decreased at 72 h. The HER- 2 protein contents of SKBR- 3 cells in the Herceptin group and Group L were decreased when compared with those in the blank control group, differences being significant(P<0.01). The HER- 2 protein contents in Group M and Group H were higher than those in the Herceptin group, differences being significant(P<0.01). There was no significant difference being found in the comparison of HER- 2 protein content between the Herceptin group and Group L(P>0.05). Conclusion:The medicated serum containing Fuzheng Xiaoliu granules can obviously inhibit the HER-2 mRNA and protein expressions in SKBR-3 cells of breast cancer,whose inhibitory effect is worse than that of Herceptin,and the medicated serum with low-dose Fuzheng Xiaoliu granules has better effect.
