槐定碱上调miR-345-5p 对乳腺癌细胞BT549 增殖、迁移和侵袭的影响

    目的:探讨槐定碱对乳腺癌细胞BT549 增殖、迁移、侵袭的影响和分子机制。方法:运用细胞计数试剂盒(CCK-8)、Transwell 实验分别检测不同浓度(1.0、2.0、4.0、8.0 mg/mL) 的槐定碱对乳腺癌细胞BT549 增殖、迁移、侵袭的影响,筛选槐定碱最佳使用浓度。将细胞BT549 分为对照(NC) 组、槐定碱组、anti-miR-NC 组、anti-miR-345-5p 组、槐定碱+anti-miR-NC组、槐定碱+anti-miR-345-5p 组。实时荧光定量聚合酶链式反应(RT-qPCR) 检测miR-345-5p 表达,检测细胞活力、迁移和侵袭能力变化。免疫印迹法(Western blot) 检测磷酸化的核转录因子(NF-κB) p65(p-p65) 和磷酸化NF-κB 抑制蛋白(p-IκBα)表达。结果:不同浓度槐定碱干预后细胞BT549 活力、迁移和侵袭细胞数均显著降低,选择4.0 mg/mL 槐定碱浓度进行后续实验。与NC 组比较,槐定碱组细胞中miR-345-5p 表达显著升高,p-p65 和p-IκBα 表达降低(P<0.05)。与anti-miR-NC 组比较,anti-miR-345-5p 组细胞活力、迁移和侵袭细胞数升高,p-p65 和p-IκBα 表达升高(P<0.05)。与槐定碱+anti-miR-NC 组比较,槐定碱+anti-miR-345-5p 组细胞活力、迁移和侵袭细胞数升高,p-p65 和p-IκBα 表达升高(P<0.05)。结论:槐定碱通过上调miR-345-5p 可抑制乳腺癌细胞BT549 增殖、迁移和侵袭,其机制可能与抑制NF-κB 信号通路有关。

    Abstract:

    Abstract:Objective:To investigate the effects of sophorindin on proliferation,migration and invasion of breast cancer cell BT549 and its molecular mechanism. Methods: The effects of different concentrations(1.0, 2.0, 4.0, 8.0 mg/mL) of sophorindin on proliferation,migration and invasion of breast cancer cell BT549 were detected by cell counting Kit(CCK- 8) and Transwell test. cells BT549 were divided into control(NC) group,sophoridine group,anti- miR- NC group,anti- miR- 345-5p group,sophoridine+anti-miR-NC group and sophoridine+anti-miR-345-5p group.The expression of miR-345-5p was detected by real- time fluorescence quantitative polymerase chain reaction(RT- qPCR) , and the changes of cell viability , migration and invasion were detected. The phosphorylated nuclear transcription factor(NF- κB) P65(p- p65) and phosphorylated NF- κB inhibitor protein(p- I κB α) were detected by Western blot. Results: After the intervention of sophoridine at different concentrations,the viability,migration and number of invasive cells of BT549 cells were significantly reduced. The concentration of sophoridine at 4.0 mg/mL was selected for follow-up experiment. Compared with NC group, the expression of miR- 345- 5p,p- p65 and p- IκBα in sophoridine group were significantly increased compared with antimiR- NC group,anti-miR-345-5p group increased cell viability,migration and invasion,p-p65 and p-IκBα compared with sophoridine+anti-miR-NC group, sophoridine+anti-miR-345-5p group increased cell viability, migration and invasion, p-p65 and p-IκBα expression increased(P<0.05). Conclusion:Sophorindin can inhibit proliferation,migration and invasion of breast cancer cell BT549 through upregulation of miR- 345- 5p, and its mechanism may be related to inhibiting NF- κB signal pathway.
