    摘要:

    目的:采用网络药理学方法探讨四君子汤治疗克罗恩病的作用靶点和信号通路。方法:通过美国国家生物技术信息中心(NCBI) 预测克罗恩病的疾病靶点,中药系统药理学数据库与分析平台(TCMSP) 筛选四君子汤的主要活性成分和靶点信息。采用Cytoscape 3.7.2 软件Bisogenet 包构建蛋白质相互作用(PPI) 网络,ClusterProfiler R 包进行靶点基因本体(GO) 分析和京都基因与基因组百科全书(KEGG) 通路富集分析。结果:预测灌木远志酮A、3β-乙酰氧基苍术酮、常春藤素、槲皮素及山奈酚等98 个活性成分,主要作用于T 细胞表面抗原CD26(DPP4)、一氧化氮合酶2 (NOS2) 及过氧化物酶体增生激活受体γ(PPARG) 等27 个克罗恩病靶点基因,通过白细胞介素(IL) -17 信号通路、肿瘤坏死因子信号通路及利什曼病等24 条信号通路发挥治疗作用。结论:四君子汤是通过多途径、多环节、多靶标发挥治疗克罗恩病的作用。

    Abstract:

    Abstract: Objective: To explore the target and signal pathway of Sijunzi decoction in the treatment of Crohn disease. Methods:The disease target of Crohn disease was predicted by the National Biotechnology Information Center(NCBI),and the main active components and target information of Sijunzi decoction were screened by TCMSP. Protein protein interaction (PPI) network was constructed by Cytoscape 3.7.2 software Bisogenet package,and ClusterProfiler R package was used for target gene ontology(GO) analysis and Kyoto Encyclopedia of genes and genomes(KEGG) pathway enrichment analysis. Results:It was predicted that frutinone A and 3β-acetoxyatractylolone,hederagenin,quercetin,kaempferol and other 98 active components mainly act on T cell surface antigen CD26(DPP4), nitric oxide synthase 2(NOS2) and peroxisome proliferation activated receptor γ(PPARG) and other 27 Crohn disease target genes play a therapeutic role through 24 signal pathways such as interleukin(IL)- 17 signal pathway, tumor necrosis factor signal pathway and Leishmaniasis. Conclusion: Sijunzi decoction plays a role in the treatment of Crohn disease through multiple channels,links and targets.
