    摘要:

    目的:观察西黄丸含药血清对人甲状腺癌细胞TPC-1 增殖、凋亡及迁移的影响。方法:SPF 级新西兰大白兔灌胃4、8、16 g/kg 西黄丸制备低、中、高浓度西黄丸含药血清,并制备对照血清,分别处理人甲状腺癌细胞株TPC-1。取对数期生长的TPC-1 细胞,分为低、中、高浓度组和对照组,每组5 个复孔,分别用低、中、高浓度西黄丸含药血清和对照血清进行干预。采用四唑盐比色法(MTT) 检测干预24h、48h、72 h 细胞增殖情况;分别采用流式细胞仪、Transwell 小室实验检测干预48 h 后细胞凋亡率、穿膜细胞数;采用实时荧光定量-聚合酶链反应法检测增殖细胞核抗原(PCNA)、细胞黏附分子1(CADM1)、B 淋巴细胞瘤-2 基因(Bcl-2)、Bcl-2 相关X 蛋白(Bax) mRNA;采用蛋白质印迹法检测PCNA、CADM1、Bcl-2、Bax 及裂解的含半胱氨酸天冬氨酸蛋白水解酶-3(Cleaved Caspase-3) 蛋白表达情况。结果:与对照组比较,低、中、高浓度组不同时间点MTT 实验OD 值均较低(P<0.05);与低浓度组比较,中浓度组48 h、72 h MTT 实验OD 值较低(P<0.05),高浓度组不同时间点MTT 实验OD 值均较低(P<0.05);与中浓度组比较,高浓度组不同时间点MTT 实验OD 值均较低(P<0.05)。与对照组比较,低、中、高浓度组细胞凋亡率及穿膜细胞数明显升高(P<0.05),且呈浓度依赖性(P<0.05)。与对照组比较,低、中、高浓度组PCNA、Bcl-2 mRNA 和蛋白相对表达量较低(P<0.05),Cleaved Caspase-3、CADM1、Bax mRNA 和蛋白相对表达量较高(P<0.05);与低浓度组比较,中、高浓度组PCNA、Bcl-2 mRNA 和蛋白相对表达量较低(P<0.05),Cleaved Caspase-3、CADM1、Bax mRNA 和蛋白相对表达量较高(P<0.05);与中浓度组比较,高浓度组PCNA、Bcl-2 mRNA 和蛋白相对表达量较低(P<0.05),Cleaved Caspase-3、CADM1、Bax mRNA 和蛋白相对表达量较高(P<0.05)。结论:西黄丸含药血清可抑制人甲状腺癌细胞株TPC-1 增殖和迁移,并促进凋亡,其中高浓度组西黄丸制备的血清效果最佳。该机制可能与下调PCNA、Bcl-2mRNA 和蛋白,上调CADM1、Bax mRNA 和蛋白及Cleaved Caspase-3 蛋白表达有关。

    Abstract:

    Abstract: Objective: To observe the effect of medicated serum of Xihuang pills on the proliferation, apoptosis and migration of human thyroid cancer cell TPC- 1. Methods: SPF New Zealand white rabbits were given 4, 8 and 16 g/kg of Xihuang pills for gastric irrigation to prepare medicated serum of Xihuang pills with low, medium and high- concentration, and the control serum was prepared,so as to treat human thyroid cancer cell line TPC-1 respectively. TPC-1 cells growing at logarithmic phase were selected and divided into the low, medium and high- concentration groups and the control group, with five replications in each group;the medicated serum of Xihuang pills with low,medium and high-concentration and the control serum were applied respectively for intervention. Methyl thiazolyl tetrazolium(MTT) was used to detect the proliferation of cells after intervention for 24, 48 and 72 hours respectively; flow cytometry and Transwell chamber experiment were used respectively to detect the apoptosis rate and the number of transmembrane cells after 48 hours of intervention;realtime fluorescence quantitative-polymerase chain reaction method was used to detect the mRNA of proliferating cell nuclear antigen(PCNA),cell adhesion molecule 1(CADM1),B-cell lymphoma- 2 gene(Bcl- 2) and Bcl- 2 associated X protein(Bax); western blotting was used to detect the expression levels of PCNA,CADM1,Bcl-2,Bax,and Cleaved Caspase-3 protein. Results:The OD values of MTT experiment at different time points in the low,medium and high-concentration groups were lower than those in the control group(P<0.05); the OD values of MTT experiment in 48 and 72 hours in the mediumconcentration group were lower than those in the low-concentration group(P<0.05),and the OD values of MTT experiment at different time points in the high-concentration group were lower(P<0.05);the OD values of MTT experiment at different time points in the high- concentration group were lower than those in the medium- concentration group(P<0.05). The apoptosis rates and the numbers of transmembrane cells in the low, medium and high- concentration groups were significantly increased when compared with those in the control group(P<0.05),with a concentration-dependent manner(P< 0.05). The relative expression levels of PCNA and Bcl-2 mRNA,and protein were lower in the groups of low,medium and high- concentration than in the control group(P<0.05), and the relative expression levels of Cleaved Caspase- 3 protein, CADM1,Bax mRNA and protein were higher(P<0.05);the relative expression levels of PCNA and Bcl-2 mRNA and protein in the medium and high- concentration groups were lower than those in the low- concentration group(P<0.05), and the relative expression levels of Cleaved Caspase- 3 protein, CADM1, Bax mRNA and protein were higher(P<0.05); the relative expression levels of PCNA and Bcl-2 mRNA and protein in the high-concentration group were lower than those in the medium- concentration group(P<0.05), and the relative expression levels of Cleaved Caspase- 3 protein, CADM1, Bax mRNA and protein were higher(P<0.05). Conclusion:The medicated serum of Xihuang pills can inhibit the proliferation and migration of human thyroid cancer cell line TPC-1 and promote the apoptosis,among which the serum prepared with highconcentration Xihuang pills has the best effect. The mechanism may be related to down-regulation of PCNA and Bcl-2 mRNA and protein,up-regulation of CADM1 and Bax mRNA and protein,and Cleaved Caspase-3 protein expression.
