    摘要:

    目的:探讨芍药甘草复合精油(SGO) 对过氧化氢(H2O2) 诱导鼠嗜铬细胞瘤(PC12) 细胞氧化 损伤的保护机制。方法:采用200 μmol/L 的H2O2 处理PC12 细胞建立氧化应激损伤模型。将PC12 细胞分为对 照组、模型组(200 μmol/L H2O2)、SGO 低浓度组(1 μmol/L SGO+200 μmol/L H2O2)、SGO 高浓度组(10 μmol/L SGO+200 μmol/L H2O2)。细胞处理结束后,采用CCK-8 法检测细胞存活率;试剂盒检测细胞超氧化物歧化 酶(SOD) 活力及丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-PX) 含量;流式细胞术检测细胞凋亡率;免 疫印迹法检测细胞中剪切化半胱氨酸天冬氨酸蛋白激酶-3 (C-Caspase3)、B 淋巴细胞瘤-2 基因(Bcl-2)、 Bcl-2 相关X 蛋白(Bax) 蛋白表达。结果:与对照组比较,模型组的细胞存活率、GSH-PX 含量、SOD 活力 及Bcl-2 蛋白表达水平明显降低(P<0.05),细胞凋亡率、MDA 含量及C-Caspase3、Bax 蛋白表达水平明显升 高(P<0.05);与模型组比较,SGO 低高浓度组的细胞凋亡率、MDA 含量及C-Caspase3、Bax 蛋白表达水平 明显降低(P<0.05),细胞存活率、GSH-PX 含量、SOD 活力及Bcl-2 蛋白表达水平明显升高(P<0.05)。结 论:SGO 对H2O2诱导的PC12 细胞氧化损伤具有保护作用,其作用机制与抗氧化应激损伤、抗细胞凋亡有关。

    Abstract:

    Abstract: Objective: To discuss the mechanism of Shaoyao Gancao Oil(SGO) protecting pheochromocytoma(PC12) cells of rats from oxidative damage induced by hydrogen peroxide(H2O2). Methods:The PC12 cells were treated with 200 μmol/L H2O2 and established as the models with oxidative damage. The PC12 cells were divided into the control group,the model group(200 μmol/L H2O2),the lowconcentration SGO group(1 μmol/L SGO+200 μmol/L H2O2) , and the high-concentration SGO group (10 μmol/L SGO+200 μmol/L H2O2). After the treatment of the PC12 cells,the cell viability was detected by the method of CCK- 8; the activity of superoxide dismutase(SOD), as well as the contents of malonic dialdehyde(MDA) and glutathione peroxidase(GSH- PX) of cells were detected by kits;the apoptosis ratio was detected by flow cytometry; the protein expressions of cleaved- caspase3(C- Caspase3), B- cell lymphoma- 2(Bcl- 2) and Bcl- 2 associated X protein(Bax) were detected by Western blot. Results: Compared with those in the control group, the cell viability, GSH- PX content, SOD activity and Bcl- 2 protein expression level in the model group were significantly decreased(P<0.05),and the apoptosis ratio, MDA content as well as the protein expression levels of C-Caspase3 and Bax were significantly increased (P<0.05); in the low- concentration SGO group and the high- concentration SGO group, the apoptosis ratios,MDA contents as well as the protein expression levels of C- Caspase3 and Bax were significantly decreased(P<0.05), and the protein expression levels of cell viability, GSH- PX contents, SOD activity and Bcl- 2 were significantly increased when compared with those in the model group respectively(P< 0.05). Conclusion:SGO has a protective effect on H2O2-inducible oxidative damage of PC12 cells,whose mechanism is related to antioxidative stress damage and apoptosis.
