    目的:观察电针对东莨菪碱诱导谵妄大鼠远期认知功能障碍的影响及可能的作用机制。方法:将 30 只SD 大鼠按随机数字表法随机分为空白组、模型组与电针组各10 只,除空白组外,其余2 组腹腔注射东 莨菪碱建立谵妄模型,电针组建模后电针神庭、印堂穴,连续7 d,7 d 后对3 组大鼠进行Y 型迷宫测试,测 试结束后取大鼠脑海马组织,用酶联免疫吸附法检测肿瘤坏死因子-α(TNF-α) 和白细胞介素-1β(IL-1β) 浓度,用TUNEL 法检测神经细胞凋亡,用蛋白质印迹法检测天冬氨酸特异性半胱氨酸蛋白酶-3(Caspase-3)、 B 淋巴细胞瘤-2 (Bcl-2) 表达。结果:与空白组比较,模型组大鼠Y 型迷宫测试总训练次数增加(P< 0.05),达标时间延长(P<0.05);与模型组比较,电针组大鼠Y 型迷宫测试总训练次数减少(P<0.05),达 标时间缩短(P<0.05)。与空白组比较,模型组大鼠海马组织TNF-α、IL-1β 水平均升高(P<0.05);与模型 组比较,电针组大鼠海马组织TNF-α、IL-1β 水平均降低(P<0.05)。与空白组比较,模型组海马组织棕黄色 颗粒明显增多,提示海马组织神经细胞凋亡增多,而电针组海马组织棕黄色颗粒较模型组明显减少,提示电针 干预可抑制东莨菪碱诱导的神经细胞凋亡。与空白组比较,模型组海马组织Capase-3 蛋白相对表达水平升 高(P<0.05),Bcl-2 蛋白相对表达水平降低(P<0.05);与模型组比较,电针组Capase-3 蛋白相对表达水平 降低(P<0.05),Bcl-2 蛋白相对表达水平升高(P<0.05)。结论:电针神庭、印堂穴可改善东莨菪碱诱导谵 妄大鼠远期认知功能障碍,其机制可能是减轻中枢炎症反应,逆转凋亡蛋白异常表达,进而抑制神经细胞 凋亡。

    Abstract:

    Abstract:Objective:To observe the effect of electro-acupuncture on long-term cognitive dysfunction in delirium rats induced by scopolamine and its possible mechanism. Methods:A total of 30 SD rats were divided into the blank group , the model group and the electro- acupuncture group according to the random number table method,with 10 rats in each group. Except for those in the blank group,the other two groups were given intraperitoneal injection with scopolamine to establish delirium models. After establishment , rats in the electro-acupuncture group was given electro-acupuncture at Shenting point (GV24) and Yintang point(GV29) for seven consecutive days. After seven days,Y-maze was conducted on rats in the three groups. After the test, the hippocampus of rats was taken, the concentration of tumor necrosis factor- α(TNF- α) and interleukin- 1β(IL- 1β) was detected by enzyme- linked immunosorbent assay,the apoptosis of nerve cells was detected by the TUNEL method,and the expressions of cysteinyl aspartate specific proteinase- 3(Caspase- 3) and B- cell lymphoma- 2(Bcl- 2) were detected by Western blot. Results:Compared with those in the blank group,the total training times of Y- maze for rats in the model group were increased(P<0.05), and the time of reaching standard was prolonged(P<0.05); compared with those in the model group, the total training times of Y- maze for rats in the electroacupuncture group were decreased(P<0.05),and the time of reaching standard was shortened(P<0.05). Compared with those in the blank group, the levels of TNF- α and IL- 1β in hippocampus in rats in the model group were increased(P<0.05);compared with those in the model group,the levels of TNF-α and IL- 1β in hippocampus in rats in the electro- acupuncture group were decreased(P<0.05). Compared with those in the blank group, the brown granules in the hippo campus in the model group were significantly increased,indicating that the apoptosis of nerve cells in the hippocampus was increased;compared with those in the model group,the brown granules in the hippo campus in the electro-acupuncture group were significantly decreased, indicating that the electroacupuncture intervention can inhibit the apoptosis of nerve cells induced by scopolamine. Compared with that in the blank group,the expression of Capase- 3 protein in the hippocampus in the model group was increased(P<0.05), and the expression of Bcl- 2 protein was decreased(P<0.05);compared with those in the model group,the expression of Capase- 3 protein in the hippocampus in the electro-acupuncture group was decreased(P<0.05),and the expression of Bcl- 2 protein was increased(P<0.05). Conclusion:Electro- acupuncture at Shenting point and Yintang point can improve cognitive dysfunction in delirium rats induced by scopolamine,whose mechanism may be to reduce the central inflammatory responses,reverse the abnormal expression of apoptotic proteins, and then inhibit the apoptosis of nerve cells.
