目的:基于mTOR 信号通路探究葛根素对慢性术后痛的影响及其作用机制。方法:50 只大鼠随机 分成对照组、假手术组、模型组、葛根素组和抑制剂组,采用大鼠皮肤/肌肉切口牵拉(SMIR) 的慢性疼痛模 型,葛根素组连续7 d 腹腔注射葛根素[100 mg/(kg·d)],抑制剂组连续 7 d 腹腔注射雷帕霉素[1 mg(/ kg·d)], 观察镇痛效果。采用机械性刺激缩爪阈值(MWT) 作为痛阈检测指标检测各组大鼠痛阈变化。取各组大鼠第 7 天的脊髓标本,用Western blot 方法检测脊髓组织mTOR、p-mTOR 的蛋白水平。免疫荧光染色法探究mTOR 在脊髓细胞的定位及观察脊髓背角小胶质细胞的激活情况。结果:葛根素、雷帕霉素可以缓解SMIR 引起的痛 觉过敏,葛根素对脊髓组织中mTOR 的表达无影响,但降低了脊髓组织中p-mTOR 的蛋白表达水平,mTOR 在脊髓背角与小胶质细胞特异性标记物共定位,葛根素、雷帕霉素可以抑制脊髓小胶质细胞的激活。结论:葛 根素可通过抑制mTOR 的磷酸化减轻小胶质细胞的激活治疗慢性术后痛。
Abstract:
Abstract:Objective:To explore the effect of puerarin on chronic postoperative pain and its mechanism based on mTOR signal pathway. Methods: Fifty rats were randomly divided into control group, shamoperation group, model group, puerarin group and inhibitor group. The chronic pain model of skin/ muscle incision and retraction (SMIR) was used in rats. Puerarin group was injected intraperitoneally with puerarin [100 mg/(kg · d)] for 7 consecutive days, and inhibitor group was injected intraperitoneally with rapamycin [1 mg/(kg · d)] for 7 consecutive days,and the analgesic effect was observed. The change of pain threshold of rats in each group was detected by mechanical withdrawal threshold (MWT). The spinal cord samples of rats in each group on the seventh day were taken, and the protein levels of mTOR and p- mTOR in spinal cord tissue were detected by Western blot. Immunofluorescence staining was used to investigate the location of mTOR in spinal cord cells and observe the activation of microglia in the dorsal horn of the spinal cord. Results:Puerarin and rapamycin can alleviate the hyperalgesia caused by SMIR. Puerarin has no effect on the expression of mTOR in the spinal cord tissue, but reduces the protein expression level of p-mTOR in the spinal cord tissue. mTOR is co-located with microglia specific markers in the dorsal horn of the spinal cord. Puerarin and rapamycin can inhibit the activation of microglia in the spinal cord. Conclusion:Puerarin can alleviate the activation of microglia by inhibiting the phosphorylation of mTOR to treat chronic postoperative pain.
