Abstract:Objective:To observe the effect of Tenglong Buzhong Granules on the metastasis of CT26 colorectal cancer. Methods:CT26 cells were injected into the tail vein of BALB/c mice. They were randomly divided into control group, small dose and large dose groups of Tenglong Buzhong Granules. They were given sterile water or small dose and large dose of Tenglong Buzhong Granules by gavage once a day for 21 days. After treatment,taking the lung to weigh the mass and count the metastatic nodules on the lung surface. Hematoxylin- eosin staining (HE) was used to observe the metastasis. Western blot was used to detect the protein expression. Results: Compared with the control group, after the treatment with Tenglong Buzhong Granules,the number of metastatic nodules on the lung surface and the quality of lung in the large and small dose groups were decreased (P<0.01). Compared with the control group, the expressions of matrix metalloproteinase-2 (MMP-2) and MMP-9 in the large and small dose groups were down-regulated (P<0.01). Compared with the control group,the expressions of Wnt protein 3 (Wnt3) and CTNNB1 protein in the large and small dose groups were down-regulated (P<0.01). Conclusion:Tenglong Buzhong Granules can inhibit the metastasis of CT26 colorectal cancer,and its mechanism may be related to the inhibition of Wnt/β-catenin signal transduction and decreased expressions of MMP-2 and MMP-9.
