    目的:研究紫菟二仙汤对雷公藤多苷致早发性卵巢功能不全模型大鼠卵巢凋亡蛋白的影响。方 法:将60 只SD 大鼠分为空白组(N 组)10 只和造模组50 只,采用雷公藤多苷灌胃法制作早发性卵巢功能不全大 鼠模型,造模成功后将造模组分为模型组(M 组)、西药组(X 组)、紫菟二仙汤低剂量组(L 组)、中剂量组(Z 组) 和高剂量组(H 组) 各10 只。给药期间观察各组大鼠动情周期,9 周后观察子宫指数及卵巢指数变化, 蛋白印迹法检测B 淋巴细胞瘤-2 (Bcl-2)、Bcl-2 相关X 蛋白(Bax)、冬氨酸特异性半胱氨酸蛋白酶- 3(Caspase-3) 蛋白含量,并计算Bcl-2/Bax 比值。结果:各组大鼠子宫指数及卵巢指数比较,差异无统计学 意义(P>0.05)。与N 组比较,M 组大鼠动情周期延长(P<0.05);与M 组比较,X、Z 及H 组大鼠动情周期 缩短(P<0.05);各治疗组动情周期比较,差异均无统计学意义(P>0.05)。各组大鼠Bax 蛋白表达水平比 较,差异均无统计学意义(P>0.05)。与N 组比较,M 组Bcl-2 表达水平及Bcl-2/Bax 降低(P<0.05), Caspase-3 表达水平升高(P<0.05);与M 组比较,X 组、Z 组及H 组Bcl-2 表达水平及Bcl-2/Bax 升高(P< 0.05),Caspase-3 表达水平下降(P<0.05),L 组Caspase-3 表达水平下降(P<0.05);与X 组比较,L 组 Bcl-2 表达水平及Bcl-2/Bax 降低(P<0.05);与L 组比较,Z 组及H 组Bcl-2 表达水平及Bcl-2/Bax 上 升(P<0.05),Caspase-3 表达水平下降(P<0.05)。结论:紫菟二仙汤可调节雷公藤多苷致早发性卵巢功能 不全模型大鼠的动情周期,其机制可能与提高Bcl-2 表达,降低Caspase-3 表达,提高Bcl-2/Bax 密切相关。

    Abstract:

    Abstract:Objective:To study the effect of Zitu Erxian Decoction on ovarian apoptosis related-proteins in model rats with premature ovarian insufficiency induced by tripterygium glycosides. Methods:Sixty SD rats were divided into blank group(group N) with 10 rats and model group with 50 rats. Rat model of premature ovarian insufficiency was established by gavage of tripterygium glycosides. After two weeks of modeling,the model group rats were divided into the model group (Group M),the western medicine group (group X),the low-dose Zitu Erxian Decoction group (Group L),the medium-dose Zitu Erxian Decoction group (Group Z), and the high- dose Zitu Erxian Decoction group (Group H), with 10 rats in each group. During the administration period, the estrous cycles of rats in each group were observed. After nine weeks,the changes in uterine indexes and ovarian indexes were observed , western Blot was used to detect the protein contents of B- cell lymphoma 2 (Bcl- 2), Bcl- 2- associated X protein (Bax), and cysteinyl aspartate specific proteinase- 3 ( Caspase- 3),and the Bcl- 2/Bax ratio was calculated. Results: There was no significant difference being found in the comparison of the uterine indexes and ovarian indexes of rats among the groups (P>0.05). Compared with that in Group N,the estrous cycle in rats in Group M was prolonged (P<0.05). Compared with that in Group M,the estrous cycles of rats in Groups X, Z,and H were shortened (P<0.05). There was no significant difference being found in the comparison of estrus cycles among the groups (P>0.05). There was no significant difference being found in the comparison of the protein expression levels of Bax among different groups of rats (P>0.05). Compared with those in Group N,the expression levels of Bcl- 2 and Bcl- 2/Bax in Group M were decreased (P<0.05), and the expression level of Caspase- 3 was increased (P<0.05). Compared with those in Group M, the expression levels of Bcl- 2 and Bcl- 2/Bax in Group X, Z, and H were increased (P<0.05), and the expression levels of Caspase- 3 were decreased (P<0.05), while the expression level of Caspase- 3 in Group L was decreased (P<0.05);compared with those in Group X,the expression levels of Bcl- 2 and Bcl-2/Bax in Group L were decreased (P<0.05);compared with those in Group L,the expression levels of Bcl-2 and Bcl-2/Bax in Group Z and H were increased (P<0.05),and the expression levels of Caspase-3 were decreased (P<0.05). Conclusion:Zitu Erxian Decoction can regulate the estrous cycles of model rats with premature ovarian insufficiency induced by tripterygium glycosides,and its mechanism may be closely related to increasing Bcl-2 expression,reducing Caspase-3 expression,and enhancing Bcl-2/Bax.
