Exploration of the Mechanism of Fuganning in the Treatment of Recurrent Respiratory Tract Infections in Children Based on Bioinformatics and Network Pharmacology
Abstract:Objective:To explore the pharmacological mechanism of Fuganning,a clinical trial recipe of Professor LI Yirui,a famous veteran doctor of traditional Chinese medicine,in the treatment of recurrent respiratory tract infections (RRTIs) in children. Methods:Using bioinformatics methods to screen the active ingredients of Chinese medicine contained in Fuganning and predict its target points of action. Screening specific gene targets for RRTIs through gene expression matrix and clinical information, intersecting Chinese medicine targets with RRTIs targets, and obtaining potential therapeutic targets for the recipe. Subsequently, therapeutic targets were imported into the STRING database to obtain protein interaction relationships, and Hub targets were screened using Cytoscape. Calculate the frequency of interactions between active compounds and Hub targets,and select the compound with the most interactions with the core target as the core compound. Finally, conduct GO and KEGG enrichment analysis and visualization. Results:This study screened a total of 139 effective active ingredients and 565 target points of Fuganning Recipe. A total of 999 differentially expressed genes in RRTIs were screened using bioinformatics methods,including 165 up-regulated differentially expressed genes and 834 down-regulated differentially expressed genes. A total of 27 therapeutic targets were obtained,of which 5 Hub targets were identified as key therapeutic targets, namely NFKBIA, JUN, TLR4, IKBKB, and CXCL8. Screen out the 10 core compounds with the most interactions with Hub targets. Enrichment analysis revealed 215 pathways, mostly related to inflammation and immunity , such as T cell receptor signaling pathways and NF- κB signaling pathway and B cell receptor signaling pathway. Conclusion:Fuganning can act on multiple targets with NFKBIA,JUN,TLR4,IKBKB,and CXCL8 as the core through active ingredients such as Quercetin, Anserine,and 3,4-O-isopropylidene shikimic acid,thereby regulating patient immune function,suppressing excessive inflammatory response,and preventing and treating pediatric RRTIs.
