    目的:观察雷公藤红素苷(Cel) 对子痫前期(PE) 大鼠肝肾细胞凋亡的影响及其保护作用。方 法:将50 只妊娠SD 大鼠随机分为对照(Control) 组、PE 组、PE+低剂量Cel (PE+L-Cel) 组、PE+中剂量 Cel (PE+M-Cel) 组、PE+高剂量Cel (PE+H-Cel) 组5 组各10 只。通过尾静脉注射脂多糖诱导PE 大鼠模 型,从孕第14 天到孕第20 天,PE+L-Cel 组、PE+M-Cel 组和PE+H-Cel 组大鼠每天分别腹腔注射5、10、 20 mg/(kg · d) 的Cel,Control 组和PE 组注射等体积的0.9%氯化钠溶液。治疗结束后,分别检测各组大鼠 的收缩压(SBP) 和尿白蛋白。通过脱氧核糖苷酸末端转移酶介导的缺口末端标记法(TUNEL) 检测肝、肾组 织细胞凋亡。通过免疫组织化学染色检测肝、肾组织中B 淋巴细胞瘤-2 基因(Bcl-2)、活化半胱胺酸蛋白酶 蛋白-9 抗体(Caspase-9) 的表达。采用酶联免疫吸附法(ELISA) 检测大鼠血清干扰素-γ(IFN-γ)、白细胞 介素-4(IL-4)、白细胞介素-17(IL-17) 和白细胞介素-10(IL-10) 水平。采用流式细胞仪检测外周血辅助 性T 细胞1(Th1)、辅助性T 细胞2(Th2)、辅助性T 细胞17(Th17)、调节性T 细胞(Treg) 占CD4+的百分 比。结果:与Control 组比较,PE 组SBP、尿白蛋白水平均升高(P<0.05),胎盘组织VEGF、MMP-2、 MMP-9 蛋白表达水平均降低(P<0.05),肝、肾组织细胞凋亡率均升高(P<0.05),肝、肾组织Caspase-9 表 达均升高(P<0.05),Bcl-2 表达均降低(P<0.05),血清IFN-γ、IL-17 水平均升高(P<0.05),IL-4、IL-10 水平均降低(P<0.05),外周血Th1、Th17 占CD4+的百分比均升高(P<0.05),Th2、Treg 占CD4+的百分比均 降低(P<0.05)。与PE 组比较,PE+L-Cel 组、PE+M-Cel 组、PE+H-Cel 组SBP、尿白蛋白水平均降低(P< 0.05),胎盘组织VEGF、MMP-2、MMP-9 蛋白表达水平均升高(P<0.05),肝、肾组织细胞凋亡率均降 低(P<0.05),Caspase-9 表达均降低(P<0.05),Bcl-2 表达均逐渐升高(P<0.05),血清IFN-γ、IL-17 水 平均降低(P<0.05),IL-4、IL-10 水平均升高(P<0.05),外周血Th1、Th17 占CD4+的百分比均降低(P< 0.05),Th2、Treg 占CD4+的百分比均升高(P<0.05)。与PE+L-Cel 组比较,PE+M-Cel 组、PE+H-Cel 组 SBP、尿白蛋白水平均降低(P<0.05),胎盘组织VEGF、MMP-2、MMP-9 蛋白表达水平均升高(P<0.05), 肝、肾组织细胞凋亡率均降低(P<0.05),肝、肾组织Caspase-9 表达均升高(P<0.05),Bcl-2 表达均降 低(P<0.05),血清IFN-γ、IL-17 水平均降低(P<0.05),IL-4、IL-10 水平均升高(P<0.05),外周血 Th1、Th17 占CD4+的百分比均降低(P<0.05),Th2、Treg 占CD4+的百分比均升高(P<0.05)。与PE+M-Cel 组比较,PE+H-Cel 组SBP、尿白蛋白水平降低(P<0.05),胎盘组织VEGF、MMP-2、MMP-9 蛋白表达水平 升高(P<0.05),肝、肾组织细胞凋亡率降低(P<0.05),肝、肾组织Caspase-9 表达均升高(P<0.05), Bcl-2 表达均降低(P<0.05),血清IFN-γ、IL-17 水平均降低(P<0.05),IL-4、IL-10 水平均升高(P< 0.05),外周血Th1、Th17 占CD4+的百分比降低(P<0.05),Th2、Treg 占CD4+的百分比升高(P<0.05)。结 论:Cel 可减轻PE 大鼠症状并抑制肝肾细胞凋亡,Cel 可能通过纠正PE 大鼠Th1/Th2 及Th17/Treg 细胞失衡来 发挥治疗作用。

    Abstract:

    Abstract:Objective:To observe the effect of Celastrol (Cel) on hepatorenal cell apoptosis of rats with preeclampsia (PE) and its protective effect. Methods:A total of 50 pregnant SD rats were randomly divided into the control group,the PE group,the PE+low-dose Cel (PE+L-Cel) group,the PE+medium-dose Cel (PE+M-Cel) group and the PE+high-dose Cel (PE+H-Cel) group,with 10 rats in each group. PE rat model was induced by injection of lipopolysaccharide through tail vein,and rats in the PE+L-Cel group,the PE+ M-Cel group and the PE + H- Cel group were respectively given the intraperitoneal injection with 5, 10, and 20 mg/(kg·d) of tretinoin glycosides per day from the day 14 of gestation to the day 20,and equal volumes of 0.9% sodium chloride solution were respectively injected into the Control group and the PE group. After treatment,the systolic pressure (SBP) and urinary albumin of rats in each group were detected respectively. Using terminal- deoxynucleotidyl transferase mediated dUTP nick- end labeling (TUNEL) method, the cell apoptosis in hepatorenal tissue was detected. The expressions of B- cell lymphoma- 2 (Bcl- 2) gene and activated cysteinyl aspartate specific proteinase- 9 (Caspase- 9) antibody in hepatorenal tissues were detected by immunohistochemical staining. The levels of serum interferon- γ (IFN- γ) , interleukin-4 (IL-4),interleukin-17 (IL-17) and interleukin-10 (IL-10) of rats were detected by enzymelinked immunosorbent assay (ELISA) method. The percentage of peripheral blood T helper cell 1 (Th1), T helper cell 2 (Th2), T helper cell 17 (Th17), and regulatory T cell (Treg) of CD4 + was detected by flow cytometry. Results:Comparing with those in the Control group,the levels of SBP and urinary albumin in the PE group were increased (P<0.05),the protein expression levels of VEGF,MMP- 2 and MMP- 9 in placental tissues were decreased (P<0.05), the hepatorenal cell apoptosis rates were increased (P< 0.05), the Caspase- 9 expression levels in hepatorenal tissues were increased (P<0.05), the Bcl- 2 expression levels were decreased (P<0.05), the levels of serum IFN- γ and IL- 17 were increased (P< 0.05),the levels of IL-4 and IL-10 were decreased (P<0.05),the percentage of peripheral blood Th1 and Th17 of CD4+ was increased (P<0.05),and the percentage of Th2 and Treg of CD4+ was decreased (P< 0.05); comparing with those in the PE group, the levels of SBP and urinary albumin in the PE + L- Cel group,the PE+M-Cel group and the PE+H-Cel group were decreased (P<0.05),the protein expression levels of VEGF,MMP- 2 and MMP- 9 in placental tissues were increased (P<0.05),the hepatorenal cell apoptosis rates were decreased (P<0.05),the Caspase- 9 expression levels in hepatorenal tissues were decreased (P<0.05), the Bcl- 2 expression levels were increased (P<0.05), the levels of serum IFN- γ and IL- 17 were decreased (P<0.05), the levels of IL- 4 and IL- 10 were increased (P<0.05), the percentage of peripheral blood Th1 and Th17 of CD4+ was decreased (P<0.05),and the percentage of Th2 and Treg of CD4 + was increased (P<0.05);comparing with those in the PE + L- Cel group,the levels of SBP and urinary albumin in the PE+M-Cel group and the PE+H-Cel group were decreased (P<0.05),the protein expression levels of VEGF, MMP- 2 and MMP- 9 in placental tissues were increased (P<0.05), the hepatorenal cell apoptosis rates were decreased (P<0.05), the Caspase- 9 expression levels in hepatorenal tissues were increased (P<0.05),the Bcl-2 expression levels were decreased (P<0.05),the levels of serum IFN-γ and IL-17 were decreased (P<0.05),the levels of IL-4 and IL-10 were increased (P<0.05), the percentage of peripheral blood Th1 and Th17 of CD4 + was decreased (P<0.05), and the percentage of Th2 and Treg of CD4 + was increased (P<0.05); comparing with those in the PE + M- Cel group, the levels of SBP and urinary albumin in the PE + H- Cel group were decreased (P<0.05), the protein expression levels of VEGF, MMP- 2 and MMP- 9 in placental tissues were increased (P<0.05), the hepatorenal cell apoptosis rates were decreased (P<0.05), the Caspase- 9 expression levels in hepatorenal tissues were increased (P<0.05),the Bcl-2 expression levels were decreased (P<0.05),the levels of serum IFN-γ and IL-17 were decreased (P<0.05),the levels of IL-4 and IL-10 were increased (P<0.05), the percentage of peripheral blood Th1 and Th17 of CD4 + was decreased (P<0.05), and the percentage of Th2 and Treg of CD4+ was increased (P<0.05). Conclusion:Cel can mitigate the symptoms of PE rats and inhibit the hepatorenal cell apoptosis. Cel may exert its therapeutic effect by correcting the imbalance of Th1/Th2 and Th17/Treg in PE rats.
