Abstract: Objective: To observe the treatment effect of modified Jingui Shenqi Pills on Th1/Th2 immune imbalance in asthma rats with kidney yang deficiency through Notch signaling pathway. Methods:A total of 40 clean grade male Wistar rats were randomly divided into the normal group (the control group), the kidney yang deficiency asthma model group( the model group),the modified Jingui Shenqi Pills group( the Chinese medicine group),and the Dexamethasone group (the western medicine group), with 10 rats in each group. Except for the control group, the model group,the Chinese medicine group,and the western medicine group all used the triple method of Hydrogenated Dexamethasone+ Ovalbumin+ Aluminum Hydroxide to construct kidney yang deficiency asthma rat models; starting from the 15th day of the experiment (i. e. the first day of stimulation), the western medicine group was given Dexamethasone Tablets at a dose of 0.25 mg(/ kg·d) by gavage, the Chinese medicine group was given the corresponding decoction of modified Jingui Shenqi Pills at a dose of 2 mg(/ kg·d) by gavage,and the control group and the model group were given 2 mL of 0.9% sodium chloride solution by gavage. After 14 days of gastric lavage treatment, rat lung tissue was stained with conventional hematoxylin eosin (HE) to observe pathological changes and airway remodeling. Th1/Th2 related cytokines were measured by enzyme-linked immunosorbent assay (ELISA),and Notch1 mRNA expression in lung tissue was detected by reverse transcription polymerase chain reaction (RT-PCR). Results: Compared with the model group,the pathological damage of lung tissue in the Chinese medicine group and the western medicine group was significantly improved, and the infiltration of inflammatory cells was reduced (P<0.05); the levels of interleukin-4( IL-4),interleukin-5( IL-5),and interleukin-13( IL-13) in the bronchoalveolar lavage fluid in the Chinese medicine group and the western medicine group were decreased (P<0.05), while the levels of interleukin-12( IL-12) and interferon-γ( INF-γ) were increased( P<0.05);the expression of Notch1m RNA in lung tissue showed a significant decrease(P<0.05). Conclusion: Modified Jingui Shenqi Pills has a certain therapeutic effect on asthma rats with kidney yang deficiency, and its mechanism is related to the downregulation of Notch1 signaling pathway to regulate Th1/Th2 imbalance of asthma rats with kidney yang deficiency.
