Abstract: Objective: To observe the clinical efficacy of Nuanshui Zaotu Formula in treating non-motor symptoms of Parkinson's disease(PD)with yang deficiency transforming into wind syndrome. Methods:A prospective controlled study was conducted on 108 PD patients with non-motor symptoms of yang deficiency transforming into wind syndrome admitted to Department of Encephalopathy, The First Affiliated Hospital of Henan University of Chinese Medicine,from January 2021 to August 2024. Patients were divided into the control group and the observation group using the random number table method,with 54 cases each. Finally,50 cases in control group(4 excluded)and 52 cases in observation group(2 excluded)completed the study. The control group received conventional treatment with Levodopa and Benserazide Hydrochloride Tablets, while the observation group received additional Nuanshui Zaotu Formula. After eight weeks of treatment, clinical efficacy and traditional Chinese medicine syndrome scores were evaluated. The Unified Parkinson's Disease Rating Scale(UPDRS)and Non-Motor Symptoms Scale(NMSS)were used to assess symptom improvement. Neurotransmitter levels [5-hydroxytryptamine (5-HT), dopamine (DA), and norepinephrine(NE)] and neurotrophic factors [nerve growth factor(NGF),glial cell line-derived neurotrophic factor (GDNF),and brain-derived neurotrophic factor(BDNF)] were measured.Incidence of adverse reactions were recorded. Results:After treatment,the total effective rate was 86.54%(45/52)in the observation group and 68.00%(34/50)in the control group,the difference being significant(P<0.05). After treatment,traditional Chinese medicine syndrome scores, UPDRS scores, and NMSS scores decreased in both groups(P<0.05), with greater reduction in the observation group(P<0.05). After treatment,5-HT,DA,and NE levels increased in both groups(P<0.05),with higher levels in the observation group(P<0.05). After treatment,serum NGF,GDNF,and BDNF levels increased in both groups(P<0.05), showing greater elevation in the observation group(P<0.05). The incidence of adverse reactions was 7.69%(4/52)in the observation group and 22.00%(11/50)in the control group,the difference being significant(P<0.05). Conclusion: Nuanshui Zaotu Formula demonstrates definite efficacy in treating non-motor symptoms of PD with yang deficiency transforming into wind syndrome. It can effectively alleviate traditional Chinese medicine syndromes,improve both motor and non-motor symptoms,regulate neurotransmitter and neurotrophic factor levels,while reducing incidence of adverse reactions.
